Abstract

Rational: Despite decades of effort based on linkage analysis, candidate gene screening, genome-wide association studies (GWAS), and a variety of other strategies, it is clear from population-based and multiple-case breast cancer family studies that the majority of the familial risk of breast cancer is still unexplained. It is very likely given current data that the remaining familial aggregations of breast cancer will be explained by several genes with a spectrum of associated risks including common genetic variants with very small associated risk of breast cancer and rare genetic mutations in genes that convey an intermediate to very high risk of breast cancer.
