Development and preliminary testing of an inactivated equine adenovirus vaccine

Abstract

SUMMARY The production and testing of an inactivated equine adenovirus (EAdV) vaccine is described. The EAdV was grown in equine fetal kidney cells. The kinetics of viral inactivation, using varied concentration of β-propiolactone (βPL) and varied incubation times, were determined. Similarly, the kinetics of inactivation of EAdV by ultraviolet, using varied exposure times, were determined. From these results, it was determined that a combined procedure of incubating EAdV for 2 hours at 37 C with 0.1% βPL and exposing for 10 minutes with ultraviolet at 188 μW/cm2 would be used for vaccine production.
